    Abstract

    Water Yam (D. alata) is one of potential tubercropsas a source of non-rice food and beneficial for health. The research aims to test the content of carbohydrate and isozyme banding pattern of Local Variety Water Yam (D. alata)local variety at Karanganyar Regency. The samples was randomly took from three part of district in Karanganyar regency, that are Gondangrejo distric, Karanganyar district and Jatipuro districts. The carbohydrate content from the seven Water Yam (D. alata) local varieties were analyzed descriptively using carbohydrate by differentmethod. The data of banding pattern isozymewas analyzed using numerical Taxonomy and Multivariate Analyst System (NTSYS). The matrix data was calculated based on DICE coefficient. The clusterization was conducted by UPGMA (Unweight Pair of Group Method With Arithmatic). The research results show that the carbohydrate content in seven local varieties of D.alatafrom the lowest arelocal variety “bangkulit”12,41 %, “beras”15,44 %, “ireng”16,77%, “alas”18,865, “legi”19,17%, “ulo”27,065 and “cicing” 37,86%. The analysis of isozyme banding pattern shows esterase enzyme expresses 10 bandsand every variety has different banding pattern on the farthest similarity range 0,53 was separated in to one ungroup individual (varietybangkulit) and one bid group (variety ireng, ulo, i beras, legi, cicing and alas) and one big group (variety ireng, ulo, beras, and legi). Shikimate dehydrogenese enzyme expresses 8 bandsand every variety has different bandingpattern. On the farthest 0,55 similarity range was separated into 2 groups, the first group (variety bangkulit, legi, beras and alas) and the second group (variety ulo and cicing). On 0,71 similarity range was separated into one big group (varietybangkulit, legi, ireng and beras) and three ungroup individual (varietyalas, ulo and cicing). Both of the enzyme (esterase and shikimate dehydrogenese) show different banding pattern from each variety, shows that there is a genetics varieties between variety from the seven Ubi Kelapa (D. alata)local variety.
