    Abstract

    ABSTRACT More than 55 % human cancer was caused by p53 mutation. Recent experimental results have proven reactivation of p53 mutant by PRIMA-1. In order to describe the restoration of p53 mutant conformation into wild type-like conformation, docking study of PRIMA-1 on R273H p53 mutant has been done. However, the dynamics of interaction between PRIMA-1 and p53 mutant at each binding site were unavailable. In order to understand the dynamics, Molecular Dynamics (MD) simulations were carried out. Four MD simulations (wild type p53, R273H p53 mutant, PRIMA-1 at R273H p53 mutant on site 1 and PRIMA-1 at R273H p53 mutant on site 2), 5 ns for each system, have been performed using the same procedure. As starting coordinate, two different sites were taken from previous docking study. These two different sites were selected, based on significant difference of docking energy. Wild type p53 and R273H p53 mutant were used as references. The results reveal that the addition of PRIMA-1 may induce the conformational change on p53 mutant. The interaction of PRIMA-1 on site 1 is more stable than PRIMA-1 on site 2. This stable interaction restores the p53 mutant conformation into wild type-like conformation. Thus, the restoration of p53 mutant conformation might be started by PRIMA-1 binding on site 1. Key words: PRIMA-1, p53, Cancer, R273H mutation, Simulation, Molecular Dinamyc.
