    Abstract

    Dragon fruit is one of tropical fruit which is potential to be developed. The information of dragon fruit’s morphology and cytology character is quite little and simple so it need to do morphology and cytology analysis. The morphology and cytology analysis give the useful information to support the dragon fruit breeding program. This research aims to study the morphology character and cytology (caryotipe) of yellow skin dragon fruit (S. megalanthus). It was took sample from “Agrowisata Kusumo Wanadri” Pantai Glagah Indah, Yogyakarta. Identification of chromosome conducted at Plants Breeding Laboratory of Agriculture Faculty UNS. It was conducted from December 2006 until June 2008. The method that was used is field research survey and squash method with pretreatment in cold water for 24 hours at 5─80C, fixation with asetat glacial 45% for 2 hours 5─80C, hydrolysis with HCL 1 N for 3-4 minutes 600C and stainning with aceto-orcein 2% for 24 hours 5─80C. The morphology and cytology data was analysed and presented descriptively. The morphology of S. megalanthus is that the root has a thread shape which is colour is white yellowish with a fibrous root system. The stem is wet, slippery and angular with convect side. The fruit is oval surrounded by short thorns, fruit’s skin is yellow, and the flesh of fruit is white. S. megalanthus has a tetraploid chromosome 2n = 4X = 44 which is length around 2 + 0,098 μm to 4,75 ± 0,098 μm. The chromosome caryotipe formula of S. megalanthus 2n = 4X = 44 = 40m + 4sm.
