    Abstract

    ABSTRACT Background: Secondary hyperuricemic is a condition caused by a disruption of uric acid production and excretion in the bodies. Increased of uric acid level may have a negative impact on the organ system, particularly kidney injury. An abnormal increased of TGF-β protein due to oxidative stress reactions may be an important factor of renal dysfunction which can cause fibrosis and glomerulosclerosis. The content of vitamin C, polyphenols, and flavonoids in the red guava fruit acts as antioxidants which could be a non-pharmacological therapy to inhibits the pathogenesis of fibrosis and renal glomerulosclerosis. Objective: This study aims to analyze effects of red guava juice dose of 5 ml/Kg mice body weight/day, dose of 10 ml/Kg mice body weight/day, dose of 20 ml/Kg mice body weight/day on fibrosis, glomerulosclerosis and expression of TGF-β protein of mice’s kidney (Mus Musculus) with secondary hyperuricemic model. Method: This study used 48 mice which assigned into 6 different groups, such as normal group, negative control group, positive control group, red guava juice dose of 5 ml/Kg mice body weight/day group, red guava juice dose of 10 ml/Kg mice body weight/day group, and red guava juice dose of 20 ml/Kg mice body weight/day group. The induction of 250mg/Kg body weight/day Potassium Oxonate during 14 days were given to negative control group, positive control, and red guava juice with various dose group. Medicine intervention with allopurinol were given to positive control group and intervention group during 21 days after potassium oxonate inducted period was done. The fibrosis and glomerulosclerosis of mice kidney were measured by hematoxylin-eosin (HE) colouring method and the expression of TGF-β protein was measured by immunohistochemistry. The data were analyzed using Krusskal Wallis with p-value <0.05 considered significant. Result: The administration of red guava juice dose of 5, 10, 20 ml/Kg mice body weight/day can inhibits fibrosis (p=0.001). Dose of 20 ml/Kg mice body weight/day shown a greater effect on inhibits renal fibrosis. Effects of the administration of red guava juice dose of 5, 10, 20 ml/Kg mice body weight/day on glomerusclerosis could not be assessed due to the gromerulus damage does not occurred (p=1.000). The administration of red guava juice dose of 5, 10, 20 ml/Kg mice body weight/day can lower the expression of TGF-β protein (p=0.026). The greatest reduction of TGF-β protein expression on secondary hyperuricemic mice model occurred on dose of 20 ml/Kg mice body weight/day. Conclusion: The administration of red guava juice can inhibits renal fibrosis in secondary hyperuricemic mice model. The administration of red guava juice with dose variation has not shown effect on glomerulosclerosis. Red guava juice can lower the expression of TGF-β protein renal of secondary hyperuricemic mice model. Keywords: Fibrosis, Glomerulosclerosis, Red Guava Juice, Secondary Hyperuricemic, TGF-β.
