    Abstract

    Abstrak Pendahuluan Faktor sekresi dari sel punca telah banyak diteliti karena memiliki manfaat dapat memperbaiki kerusakan jaringan maupun organ, dan dijumpai pada media kultur sel punca sehingga disebut sebagai media terkondisi. Advanced adipose-derived stem cell protein extract (AAPE) adalah media terkondisi dari adipose derived stem cell (ADSC) mengandung sejumlah growth factor yang disekresikan ADSC, memiliki manfaat untuk pengobatan masalah kulit seperti kerutan pada wajah serta perbaikan luka. AAPE dapat merangsang sintesis kolagen dan migrasi sel fibroblas sehingga dapat digunakan untuk memperbaiki kerutan wajah dan merangsang penyembuhan luka. Metode Penelitian in vitro ini menggunakan sampel kelompok fibroblas yang di kultur dari tali pusat, diperoleh dari seorang pendonor wanita usia 30 tahun dengan riwayat seksio sesaria pertama kali. Ada 3 kelompok subyek penelitian yaitu satu adalah fibroblas normal tanpa paparan UVB sebagai kontrol, dua perlakuan fibroblas dipapar sinar UVB 30 menit dosis 100 mJ/cm2, ditambah AAPE dan hanya basal media. Semua kelompok di evaluasi 48 jam dan dilakukan pemeriksaan deposisi kolagen dengan pengecatan imunositokimia antibodi kolagen 1, selanjutnya hasil dibaca menggunakan software Image-J. Hasil Terdapat perbedaan signifikan pada peningkatan deposisi kolagen antara kelompok kontrol, basal media dan AAPE (+). Peningkatan deposisi kolagen terbukti signifikan pada kelompok fibroblas tua yang diberi penambahan AAPE dibandingkan pada kelompok yang tanpa penambahan AAPE/hanya basal media dan kelompok kontrol, dan didapatkan nilai p< 0,001. Kesimpulan AAPE dapat memperbaiki fungsi fibroblas tua yang ditandai dengan peningkatan deposisi kolagen melalui kultur fibroblas tua. Kata kunci : AAPE, deposisi kolagen, fibroblas tua ABSTRACT Introduction The secretory factors of stem cell has been investigated because of their efficacy can repair the tissue damage and organ, also can found in cultured medium of stem cell, and called with conditioned medium. Advanced Adipose- derived Stem Cell Protein Extract (AAPE) is a conditioned medium of adipose derived stem cell (ADSC) contained several growth factors which secreted by their stem cell source. These conditioned medium had benefit for skin problem such as wrinkles and wound repair. AAPE can induce the collagen synthesis in dermis and fibroblast migration so can used to repair the wrinkles and stimulate the wound healing. Methods An in vitro study was conducted by using fibroblast sample that from amnion cultured of 30 year old woman who underwent caesarian for the first time and willing to donor. The were 3 study groups, normal fibroblast or without UV exposure as control, second and third are the treatment groups contains each fibroblast that had exposed with UVB light for 30 minutes and 100 mJ/cm2 in dose, then divided in two groups that treated with AAPE and only basal media/without AAPE. All groups were evaluated in 48 hours and did assessed the collagen deposition by immunochemistry staining using collagen antibody type 1, then the results were analyzed by Image-J software. Results There were significant differences in the increased of collagen deposition between control group and study groups. UVB exposed fibroblast added with AAPE had the greatest amount of collagen deposition than control and only basal media groups, with p value 0.001. Conclusion AAPE can repair the senescent fibroblast function by increasing the collagen deposition of senescent fibroblast cultured. Key word : AAPE, collagen deposition, senescent fibroblast
