    Abstract

    ABSTRACT DINA IKA MULIAWATI. 2006. SYNTHESIS OF BIOSURFACTANTS USING SOY BEAN OIL AS ADDITIONAL CARBON SOURCE THROUGH BIOTRANSFORMATION BY Pseudomonas aeruginosa Synthesis of biosurfactants using soy bean oil as additional carbon source through biotransformation route by Pseudomonas aeruginosa had been done. Soy bean oil was used as additional carbon source because it has high amount of unsaturated fatty acid and could produce biosurfactant through biotransformation and would have own characteristic. Growth curve was made to know the optimum inoculation time of fermentation media. The production of biosurfactant was optimized at different concentration of soy bean oil e.g 0, 5, 10, 20 % (v/v) at room temperature and gyratory shaking at 150 rpm for 12 days. Every day, the sample was measured on optical density, surface tension, and emulsification index. Recovery of biosurfactant was done by extraction with increasing polarity rate of the solvent, that’s hexane, chloroform, ethyl acetate, and buthanol. Biosurfactant from the recovery process was identified using TLC and FT-IR. It was characterized emulsion type, value of CMC, and test of emulsifier for some hydrocarbon. From growth curve, optimum inoculation time was 12 hours. Addition soy bean oil as additional carbon source gave biosurfactant with better ability to reduce surface tension and make emulsion. Optimal condition of biosurfactant production was 6 days fermentation with soy bean oil concentration of 10% (v/v). Biosurfactant that accumulated in chloroform (bioSklorK) had the highest ability to decrease surface tension of palm oil and had highest emulsification index. Identification using TLC could not yet confirm the amount of compound of bioSklorK. Identification using FT-IR showed biosurfactant had hydroxyl and carbocylic as hydrophilic moieties and alyphatic long chain of hydrocarbon as hydrophobic moeities. BioSklorK had o/w type emulsion, value of CMC was 859.329 mg/L and decreased surface tension till 0.052 N/m. BioSklorK also had proved could decrease surface tension and could make emultion with palm oil, gasoline, benzene, and toluene.
