Abstract

ABSTRACT DEWI ANDRIYANI. 2005. ISOLATION AND IDENTIFICATION OF HALOPHILIC BACTERIA FROM SALTED FISH. BIOLOGY. Mathematic and Natural Science Faculty. Sebelas Maret University. Halophilic bacteria is group of microorganism that can grow in high salinity up to 30%. This bacteria could be found from salted food, e.g. salted fish. This research was done to know the species of halophilic bacteria from salted fish. Isolation of halophilic bacteria was begun from making of enriched culture. Two grams salted fish had been destroyed and mixed with 20 ml Syntetic Sea Water medium at 15% salinity (SW-15) and supplemented with 0,5% yeast extract. The enriched culture was incubated for 4 days at the room temperature and it was laid on the shaker with 120 rpm. After incubation time, the enriched culture was growth on the SW-15 with 0,5% yeast extract and 1,5% agar by spread method and dilution series. The culture was incubated for 4 days at the room temperature. The colony grow was taken and purified. Pure culture was stored at the tube with SW-15 medium, supplemented by 0,5% yeast extract and 1,5% agar. There were 28 halophilic bacterias identified with morphology, microscopies, biochemistry and physiology caracteristics and growing test on the varians of salinity. The genera of halophilic bacterias were Pseudomonas, Chromohalobacter, Halobacter, Deleya, Bacillus, Salinicoccus, Marinococcus and Kurthia. The species of halophilic bacterias are Pseudomonas sp. IVAk, Pseudomonas sp. IVAc, Pseudomonas sp. VIAa, Chromohalobacter sp. IBb, Halomonas sp. VBr22, Deleya sp. VIIBb2, Bacillus sp. IAb, Bacillus sp. IIIAc2, Bacillus sp. IIIAa, Bacillus sp. VAc1, Bacillus sp. VAa, Bacillus sp. VIAc1, Bacillus sp. IBa22, Bacillus sp. IIBb, Bacillus sp. IIIBb1, Bacillus sp. IIIBb2, Bacillus sp. IIIBa3, Bacillus sp. IIIBc1, Bacillus sp. VB21, Bacillus sp. VBb, Bacillus sp. VBc, Bacillus sp. VBr11, Bacillus sp. VIIBa2, Bacillus sp. VIIBb1, Salinicoccus sp. VBa1, Salinicoccus sp. IIIAb, Marinococcus sp. IBa21 and Kurthia sp. VIAb.
