    Abstract

    ABSTRACT THE ASSOCIATION OF GEN HUMAN LEUKOCYTE ANTIGEN POLYMORPHISMS DQB1 *0503, DQB1 * 0201, AND INTERLEUKIN-10 1082 WITH POTENTIAL EVENTS OF DRUG INDUCED LIVER INJURY IN MDR-TB PATIENTS Ari Kuncoro Tuberculosis is a disease with high mortality in adult patients. Morbidity and mortality are increasing in MDR-TB. MDR-TB treatment is more complex and requires a second-line ATD given in a long time with a risk of DILI. Gene polymorphism of class II HLA and IL-10 1082 has been known play a role in DILI events. Analytics observational with cross sectional study design in MDR-TB patients who treated at hospital Dr. Moewardi hospital Surakarta in 2011-2015. Subjects research 89 samples that met the inclusion criteria and oak sklusi premises n 47 (52.8%) men and 42 (47.2%) patients were female. Prevalensi potential incidence of DILI in MDR-TB patients withpolymorphisms of HLA-DQB1 * 0503 (20.0%) was higher than that do not have the polymorphism of HLA-DQB1 * 0503 (11.4%). However, the prevalence difference was not statistically significant (p = 0.263). DILI on the prevalence of MDR-TB patients who have polymorphisms of HLA-DQB1 *0201 (23.5%) is also greater than in MDR-TB patients who do not have the polymorphism of HLA-DQB1 *0201 (13.9%), but the statisticaltest the difference is not significant (p = 0.326). The prevalence of potential DILI incidence in patients with IL-10 1082 genotype GG polymorphism was 2 of 4 patients (50.0%), AA genotypic patients were 4 of 12 patients (33.3%), and GA genotype patients were 8 of 73 patients (11.0%). Statistical testing states that differences in the prevalence of the n pasie three groups was significant (p = 0.022). Polymorphisms of HLA-DQB1 *0503 and HLA-DQB1*0201 was not significantly associated with the potential incidence of DILI. There was a significant association between interleukin-10 1082 gene polymorphisms against potential DILI incidence in MDR-TB patients. Gene polymorphisms IL-10 1082 GA genotype have a higher protective effect than AA and GG. Keywords: multidrug-resistant TB, HLA-DQB1 gene polymorphism, IL-10, DILI.
