    Abstract

    CHARACTERIZATION AND MEASUREMENT OF STABILITY ß-GLUCOSIDASE Bacillus pumilus GOR2 FROM DIGESTION TRACT OF KUMBANG BADAK LARVAE (Oryctes rhinoceros) Robiah Anitasari Departement of Biology, Faculty of Mathematics and Natural Science Sebelas Maret University, Surakarta Cellulose is a polysaccharide found in nature. Cellulose is the primary component of plant cell wall. Cellulose can be hydrolized to simpler sugar. β-glucosidase is an important enzyme to produce glucose from cellobiose. Characterization of cellulase enzyme can help determine the optimum conditions the enzymes at work. The aims of this research were found the optimum condition of temperature, pH, and substrate concentration for activity of β-glucosidase Bacillus pumilus GOR2 from digestion tract of kumbang badak larvae (Oryctes rhinoceros), and was found determine the effect of storage time on the stability of the enzyme β-glucosidase. The bacteria used in this research was Bacillus pumilus GOR2 from digestion tract of kumbang badak larvae (Oryctes rhinoceros). β-glucosidase activity was determined using selobiose substrate. Temperature variations used were 30°C, 40°C, 50°C, 60°C, and 70°C. pH variations used were 4, 5, 6, 7, and 8. Concentration variations used were 5 mM, 10 mM, 15 mM, 20 mM and 25 mM. Measurements of stability carried out on the of incubation for 1, 2, 3, 4, and 5 hours at a temperature of 30oC, 40oC, 50oC, and 60oC and then testing activity was on the temperature, pH, and the optimum substrate concentration. The results showed optimum condition for specific activity of β-glucosidase were 50°C, pH 5, and 20 mM substrate concentration with the value of the specific activity 158.0528 U/mg protein. β-glucosidase was stable in the temperature range of 30-60°C until the long incubation time of 3 hours.
