    Abstract

    Mutamba plant (Guazuma ulmifolia Lamk.) is one of herbal plant that has been used by a lot of people in Indonesia. The main medicinal used of this plant is for slimming body. Secondary metabolites ingredients of mutamba plant which are medicinal useful can be produced rapidly by in vitro callus induction. This research was a callus induction of mutamba plant in various media composition under light and dark condition. This research was aimed to study the comparison of callus induction in various media composition under light and dark condition. The research was conducted in Plant Physiology and Biotechnology Laboratory of Agriculture Faculty Sebelas Maret University from December 2007 until March 2008, used Random Complete Design with single factor consist of 7 treatments repeated 3 times. The treatments were Gamborg (B5) + sucrose 2% + 2,4D 1 mg/l (light); Gamborg (B5) + sucrose 2% + 2,4D 1 mg/l (dark); Gamborg (B5) + sucrose 3% + 2,4D 2 mg/l (light); Gamborg (B5) + sucrose 3% + 2,4D 2 mg/l (dark); MS + sucrose 8% + 2,4D 1 mg/l (light); MS + sucrose 8% + 2,4D 1 mg/l (dark) and MS + 2,4-D 0,6 mg/l + 1 mg/l kinetin (control) The result observation data of callus initiation time, callus volume, fresh weight callus and dry weight callus variables were analyzed with variance test that separated in comparison by Contrast Test in level 5%. Contrast test among the treatment were : control with all treatments; B5 medium with MS medium; light condition with dark condition; 1 mg/l 2,4-D with 2 mg/l 2,4-D; 2 mg/l 2,4-D light condition with 2,4-D dark condition; 2% and 3% sucrose with 8% sucrose. Data of texture and colour callus variables were analyzed descriptively. Callus volume and fresh callus weight variable were also analyzed using correlation test. The result showed that callus growth in 2,4-D with same concentration in light condition was better than in dark condition. The use of 1 mg/l kinetin and 0,6 mg/l in MS media was not different in callus formation compare with media which only used 2,4-D. The treatment which showed the best callus growth was Gamborg (B5) + sucrose 3% + 2,4D 2 mg/l (light).
