      Salak pondoh is one of the salak cultivars that preffered by Indonesian people because the sweet taste even in the young fruit. Flesh of salak pondoh fruit is thin and the size of the fruit is smaller than other cultivars, these characteristic are often complained of the community, so it is necessary to improve the quality of salak pondoh with one of the plant breeding techniques that is polyploidization. This study aims to find out the proper application and concentration to produce polyploid salak. This research was conducted in greenhouse and Plant Physiology and Biotechnology Laboratory, Faculty of Agriculture, Sebelas Maret University of Surakarta in May 2016 - March 2017. This research was designed by treatments of colchicine application which consist of two, they were dripping and immersion of seed cotyledon seeds. Dripping petiole cotyledon seeds treatment consist of 3 levels concentration: control; 0.1%; and 0.2%. Seed which petiole cotyledon emerged then immersed with colchichne, which treatment consist of 2 factors: concentration, consist of 5 levels: 0.02%; 0.02%; 0.04%, 0.06%; and 0.08%. Immersion duration: 6 hours and 12 hours. The observed data were analyzed descriptively. The results showed that immersion of colchicine in all concentrations have not produced the number of polyploid chromosomes (2n = 28). Colchicine dripping in concentration of 0.02% can produce a polypoid cell, with the number of chromosomes of 56 (2n = 4x = 56).
