    Abstract

    OPTIMIZATION FORMULA OF SNEDDS (SELF-NANOEMULSIFYING DRUG DELIVERY SYSTEM) FROM EXTRACT OF CLOVE LEAF (Syzigium aromaticum L Merrill & Perry) CHLOROFORM Nyanti Muharomah Diploma 3 Pharmacy, Faculty of Mathematic and Science Sebelas Maret University ABSTRACT Clove leaf are one natural substance that believed to have many health benefits, so widely used as an alternative treatment. But the leaf extract has a low solubility and that can cause unmaximum oral bioavailability. The aim of this study is to overcome solubility of leaf extract which made by the preparation of Self-Nanoemulsifying Drug Delivery System (SNEDDS). This research was an experimental methods which carried out by extracting the leaves with maceration method using chloroform as a solvent. Tween 80 and PEG 400 are used in the formulation of SNEDDS chloroform extract of clove leaf determined by simplex lattice design. Responses were tested against 8 formula, namely the level of emulsions clarity was measured using a spectrophotometer uv/vis at a wavelength of 650 nm, emulsification time in an artificial gastric fluid and the stability SNEDDS. Optimization is done by analyzing the clarity and response time of emulsification by using Design Expert software. Result of SNEDDS optimization then characterized in diameter and size distribution after the emulsification. The maximum dose of ectract that can be incorporated in the system of SNEDDS was also tested at the optimum formula. SNEDDS produce a homogeneous emulsion and clear (clarity of 91,862 %) emulsification time of SNEDDS are 6 second with a time of 6 in artificial gastric fluid, 12 seconds to water and SNEDDS stable in room temperature. The optimum composition SNEDDS clove leaf consists of 0.71 mL of pecan oil, 3.8 gram of Tween 80 83,3 %, and 16,7 % of PEG 400 that can load 150 mg chloroform extract of clove leaf every system. The particle size of SNEDDS emulsions 17,8 nm with a uniform droplet size distribution (PI = 0.161). Keyword: Clove leaf chloroform extract, PEG 400,Tween 80, SNEDDS
