    Abstract

Mini Thesis. Faculty of Medicine, University of Sebelas Maret, Surakarta. Background: Increased activity of Adenosine Monophosphate-Activated Protein Kinase (AMPK) can inhibit cell cycle and biosynthetic pathways necessary during cancer cell growth. Adenosine Monophosphate (AMP) shows directly to activate AMPK. Therefore, it is necessary to study effectively to find a selective activator of AMPK. The aim of this study was to identify active compounds of Indonesian Herbal plants that activate AMPK by using molecular docking. We used molecular docking to help find the active compound from Indonesian herbal plants which has similarities with AMP on activating AMPK. Methods: Samples used in this study is the active compound have to meet criteria(1) listed on HerbalDB (2) has a three-dimensional structure, and (3) meet criteria Lipinski rule of five. Molecular docking is used to find active compounds have similarities with AMP on AMPK activity. This study used the program Autodock Vina version 1.1.2 as molecular docking program and Chimera 1.9 as a visualization program. Results: There were 473 active compounds derived from Indonesian herbal plants.The results of this study reported some compounds have lowerdocking scores than AMP (-7,5 Cal/mol, -8,3 Cal/mol, -8,6 Cal/mol) andcan occupy three binding site on γ subunit of AMPK. Conclusions: This study concludes that some of the compounds contained in the Indonesian medicinal plants are similar to AMP on AMPK activity because it can occupy three binding site. Hence it is thought to activate AMPK as candidate anti-cancer. Keywords: AMPK,Herbal, Molecular Docking
