    Abstract

    Effect Of L-Arginine To Endothelial SpiralisArteri Defect Of Preeclampsia-Model Mice Pengaruh L-ArginineTerhadap KerusakanEndotelArteriSpiralisMencit Model Preeklampsia Anwar Sandi Wibowo, Sri Sulistyowati, Sutrisno Department of Obstetrics and Gynecology Faculty of Medicine SebelasMaret University/ Dr. Moewardi General Hospital Surakarta Abstract Background: Preeclampsia is one of the major contributors to morbidity and mortality in the mother and fetus. Imbalance of pro-angiogenic factors and anti-angiogenic factorhas important role in pathogenesis of preeclampsia. Endothelial disfunction lead to arterispiralis defect in preeclampsia. Provision L-Arginin is suspected decreased endothelial defect. This study aims to prove the effect of L-Arginin towards improvement of endothelial dysfunction assessed by discovering the spiralisarteri diameter and thickness of preeclampsia-model mice (Musmusculus). Methods: This analytic experimental study perform in 30 health mice (Musmusculus) that pregnant for 16 days, with 20-25 gram bodyweight, to analyze ArteriSpiralis histopathology in 3 groups, that are 10 normal pregnant mice (k(-)), 10 preeclampsia mice (K(+)), 10 preeclampsia that treat by L-Arginin (P). Sampling have done in all mice that have womb surgery, continues to Hematoxylin Eosin (HE) colouring to perform histopathology procedures. Results from each sample assessed semiquantitatively according Remmele-modified methods, where the Remmele index scale (immuno reactive score / IRS) is the result of multiplying the percentage of immunoreactive cells score to the color intensity immunoreactive cells score. Data analysis was done usingKruskal Wallis and Mann Whitneyperformed by SPSS(Software Package for Social Science) 21. Results:ArteriSpiralis mean diameter of group K(-) is 25.06 + 4.94 per µm2, K(+) groups is 16.64 + 3.68 per µm2 , and (P) groups is 22.66+ 7,53 per µm2 . Mean of ArteriSpiralis thickness for K(-) groups is 53.95+ 26.96 per µm2, K(+) group is 96.50 + 16.66 per µm2, and (P) groups is 62.79+ 8.04per µm2. We found significant differences in normal groups compare to preeclampsia groups (p=0.001*), and also in preeclampsia group compare to treatment groups (p=0.000*). But in preeclampsia groups compare to normal groups there is no significant differences (p=1.000). So we can see that L-Arginin treatment is effective to repairing preeclampsia symptoms specially in ArteriSpiralis damage in preeclampsia mice models. Conclusion: There are proven significant effectiveness of L-Arginin treatment in reparing of ArteriSpiralis damage (tunika intima hyperplasia and also atherosis) in preeclampsia mice models (Musmusculus).
