    Abstract

    Emprit ginger (Zingiber officinale var. Amarum) is one type of ginger that is widely used in the food industry. One use of ginger is to perform distillation which will generate volatile oil and ginger dregs. Ginger dregs are usually only used as fuel refining. Therefore, further research on the dregs ginger terms of antimicrobial activity. This study aims to determine the effect of temperature, the effect of time and the combined effect of temperature and time and also the best of temperature and time of extraction maceration dregs ginger (Zingiber officinale var. Amarum) on microbial activity Aspergillus niger, Lactobacillus plantarum, Pseudomonas fluorescens, Staphylococcus aureus and Escherichia coli. This study uses a completely randomized design with two factors, variations in temperature (55°C; 75°C; 95°C) and a variation of the extraction time (15, 30, 45 minutes). Repetition sample 2 times to repeat with 2 times a repeat analysis. Results showed antimicrobial activity on the bacterium E. coli 104 cells/ml by the formation of inhibition zone of 17.73±0.03 mm with the temperature and the extraction time is 95°C for 45 minutes. Diameter of inhibition zone on the testing of S. aureus 103 cells/ml of 12.08±0.01 mm with a temperature of 95°C and the extraction time is 45 minutes. P. fluorescens inhibition zone at 104 cells/ml is 13.02±0.01 mm with a temperature of 95°C and the extraction time is 45 minutes. Diameter of inhibition zone in A. niger 104 cells/ml of 11.70±0.08 at a temperature of 75oC with a time of 30 minutes. In testing the L. plantarum no inhibition zone formed at all temperature treatment and extraction time. The higher temperature of extraction maceration then the antimicrobial activity of ginger dregs extract againts E. coli, S. aureus and P. fluorescens higher but do not apply the A. niger. The longer time of extraction maceration then the antimicrobial activity of ginger dregs extract againts E. coli, S. aureus and P. fluorescens higher but do not apply the A. niger. The combination of temperature and time is showing that there’s an interaction of the antimicrobial activity of ginger dregs extract on A. niger but not on other microbes.
