    Abstract

    FARADILLAH RAHMY SAVITRI, G0006076, 2010. The Extract of Nigella sativa seed antifungal effect on Microsporum gypseum in vitro. Faculty of Medicine, Sebelas Maret University, Surakarta. Objective: Nigella sativa seed contains thymoquinone, carvacrol, and thymol which has a function as an antifungal. This study aims to determine the effect of extract of Nigella sativa seed in influencing the growth of Microsporum gypseum in vitro. Methods: The study was perform as experimental laboratory. The object of the study is Microsporum gypseum which took by random sampling standardized by Mc Farland technique (equivalent with 0,5 Mc Farland turbidity). The study used Microsporum gypseum colonies on 11 Sabouraud Dextrose Agar plate which have contains cloramphenikol. Each plate has 4 holes. In every holes filled by etanol 70 % as negative control (K1), fluconazole 25 μg as negative control (K2) and various extract of Nigella sativa seed concentration K3-K7 (60 %, 65 %, 70 %, 75 %, and 80 %). The plate was incubated in 30o C incubator for 6 days and measured the diameter of inhibition zone. The data was collected and analyzed by Kruskal Wallis Test and Mann Whitney test on SPSS 16,0 for Windows. Result: The result of study showed that means of the diameter of inhibition zone K1 0 mm, K2 17 mm, K3 15,8 mm, K4 16,5 mm, K5 17,7 mm, K6 17,8 mm, K7 20,8 mm. The Kruskal wallis test showed that there was difference of the diameter of inhibition zone means between all of the group (K1-K7) significantly (p < 0,05). The Mann Whitney test showed that there was difference between negative control with the all of various extract of Nigella sativa seed (p < 0,05). The positive control’s diameter of inhibition zone compare to only 80 % extract of Nigella sativa seed concentration has significantly. Conclusion: The study was concluded that there is an antifungal effect of extract of Nigella sativa seed to Microsporum gypseum. 0,05 ml extract of Nigella sativa seed concentration 80 % for an antifungal on Microsporum gypseum in vitro has inhibition diameter much the same with 0,05 ml fluconazole consentration 2,5 x 10-5 %.
