    Abstract

    Binuangeun Beach, Banten is Java south coast which has high abundance of algae. Algae has been used extensively as medicine because its bioactive compounds that provide diverse bioactivity such as anti-viral and antioxidant. One of algal bioactive compounds which potential to be studied is lectin. Lectin is carbohydrate/glycoprotein-binding protein and plays a role in cells or cell-matrix interactions. Due to its characteristics, lectin is useful in biological research on field of glycomic and biomedic. Agal lectins has several advantages compared with higher plant lectins such as lower molecular weight, monomeric form, stable to high temperatures, and divalent cation-independent. Algal lectins don’t have an affinity for simple sugars but more specific to oligosaccharides complex, usually glycoproteins particularly animal glycoproteins. The coarse fraction of lectin on Enteromorpha clathrata has been extracted using ammonium sulfate precipitation method. Hemagglutination activity was tested with rabbit and human erythrocytes (A, B and O Type) with treatments (native and trypsin-treated). The highest activity of rabbit erythrocytes in TRBC and human erythrocytes in RBC A. The lectin coarse fraction partially purified, obtained 3 protein bands with highest molecular weight 41.82 kDa and lowest At 11.43 kDa. In carbohydrate-binding specificity test, showed no binding for monosaccharides and oligosaccharides, but inhibited by some glycoproteins with aPTG as the biggest obstacle. In stability test, the hemagglutination activity is stable at high temperature (23 to 1000C) and wide pH (pH 23 to 10) and the divalent cation- independent (23 on the addition of EDTA, CaCl2 and MgCl2).
