    Abstract

    Nifedipine is used for hypertension treatment which are included in the class of calcium channel blocker with a half-life of 4 hours. In general, hypertension treatment needed the long term. Nifedipine formulationsin sustained release dosage forms can maintain therapeutic drug levels at concentrations in the long term. This study aimed to determined the effect of chitosan concentration and effect of the method of making the sustained release nifedipine tablet with Na-montmorillonite matrix of chitosan. The process of making the tablets by direct clamp was used different chitosan concentration for each formula are 20 mg and 40 mg. The method were nanocomposite and non-nanocomposite. Weight of each tablet was 200 mg. Each formula tested by the physical properties of the tablet form contain of hardness test, swelling test and drug release test with disolution appliance tester. Dissolution test was performed for 6 hours in simulated gastric fluid medium with the addition of 0.5% sodium lauryl sulfate at 100 rpm at 37 ± 0,5˚C. The data result were compared with a reference standard and analyzed statistically by one way ANOVA with a 95% confidence interval. If there was a significant difference, LSD t-test could be continued. Results showed that the difference of chitosan concentrations and method of manufacture may affect the drug released profile and DE360 value. The increasing of chitosan concentration caused decreasing of DE360 value. Release Kinetics approached Higuchi (nanocomposite method) and order one (non-nanocomposite method), while the release mechanism in the formula 1A, 1B and 2B was not classified and formula 2A was Anomalous transport.
