    Abstract

    Nifedipine is antihypertensive drug. Sustained release tablet is suitable design for nifedipin which has a short half-life, only 2 hours. This research is aimed for optimize and assess the affect of chitosan and Carbopol®940 proportion on drug releasing profile and drug release kinetics of nifedipin sustained release tablet. Nifedipine sustained release tablet made by nanocomposite system. The 22factorial design method is applicated to optimize and assess the affect of chitosan and Carbopol®940 proportion as independet variable. Direct compression method used in tablet preparation with four formula, it’s chitosan20 mg and Carbopol®940(F1), chitosan 20 mg and Carbopol®940 30 mg(Fa), chitosan 40 mg and Carbopol®940 10 mg(Fb), Chitosan 40 mg and Carbopol®940 30 mg(Fab). The optimum area determined by desirability plot with dissolution efficiency parameter (DE). Releasement profile(Q60 minutes and Q360 minutes),index swelling and hardness use Design Expert®. The result shows that drug release kinetics of F1, Fa, Fb approach higuchi kinetics and Fab approach first order. Based on design expert, that chitosan as the most dominant factor affected on DE and drug releasing profile. The interaction of both factors affected on increasing hardness and decreasing index swelling. Based on desirability plot was obatained optimum area at chitosan factor 26,35 mg and Carbopol®940 in 30 mg. Key word: nanocomposite , factorial design, chitosan, Carbopol ®940, optimum formula
