    Abstract

    Objective: The objectives of this research are to know the influence of bayleaf extract and the increase of bayleaf extract dose on mice liver damage induced by paracetamol. Methods: This was laboratory experimental research with the post test only controlled group design. Samples were 28 male mice, Swiss Webster type, 2-3 months old age and + 20 g of each weight. Samples were divided into 4 groups of 7 mice each. This research used incidental sampling technique. Bayleaf extract was given for 14 days in a row, while paracetamol in toxic dose was given on the 12th, 13th, and 14th day. Samples were divided into 4 group : negative control group which was given aquadest only per oral everyday; positive control group which was given toxic dose paracetamol; experimental group 1 (P1) which was given 7 mg/20 g weight mice dose bay leaf extract and toxic dose paracetamol and experimental group 2 (P2) which was given 14 mg/20 g weight of mice dose bay leaf extract and toxic dose paracetamol. On the 15th day, mice were sacrificed by neck dislocation. After that, the liver was dissected out and made for preparat with paraffin block method which was stained by Hematoxillin Eosin (HE). Preparat was observed. The score of hepatocyte damage was determained by pyknosis, karyorrhexis, and karyolysis cells. The data was analized by One-Way ANOVA tes (α = 0,05) and continued by Post Hoc Multiple Comparisons using Least Significant Differences (LSD) test (α = 0,05). Results: The result of one way ANOVA test showed that there was significant differences in score of hepatocyte (p = 0,000). LSD test showed a significant differences in score of hepatocyte (p = 0,000) across all pair groups. Conclusion: Bay leaf extract can decrease the liver histological structure in mice induced by paracetamol and higher dose of bay leaf extract, higher its histology protection againts liver histological damage from paracetamol toxic on mice therefore can’t be like control group.
