    Abstract

    Wahyu Wirawan, G0010195, 2013. The Effect of Mango (Mangifera indica L.) Fruit Juice on Paracetamol – Induced Renal Histological Structure in Mice (Mus musculus). Mini Thesis. Faculty of Medicine, Sebelas Maret University, Surakarta. Objective: The objectives of this research are to know the influence of Mango fruit juice and the increase of Mango fruit juice dose on mice renal damage induced by paracetamol. Methods: This was laboratory experimental research with randomized controlled trial design. Samples were 28 male mice, Swiss webster type, 2-3 months old age and + 20 g of each weight. Samples were divided into 4 groups of 7 mice each. This research used purposive sampling technique. Mango juice was given for 14 days in a row, while paracetamol in toxic dose was given on the 12 th , 13 th , and 14 th day. Samples were divided into 4 groups : Negative Control Group (KKn) which was given aquadest only; Positive Control Group (KKp) which was given toxic dose paracetamol; Experimental Group 1 (KP1) which was given 1 st dose Mango fruit juice and toxic dose paracetamol and Experimental Group 2 (KP2) which was given 2 nd dose Mango fruit juice and toxic dose paracetamol. On the 15 th day, mice were sacrificed by neck dislocation. After that, the ren was dissected out and made for preparation with paraffin block method which was stained by Hematoxillin Eosin (HE). Preparation was observed under light microscope. The score of renal cell damage was determained by pyknosis, karyorrhexis, and karyolysis cells. The data was analized by One-Way ANOVA test (α = 0,05) and continued by Least Significant Differences (LSD) test (α = 0,05). Results: The result of One-Way ANOVA test showed that there was a significant difference in score of renal cells (p < 0,005). LSD test showed a significant difference in score of renal cells (p < 0,005) across all pair groups. Conclusion: Mango fruit juice can decrease the damage of renal histological structure in mice induced by paracetamol, and increasing the dose of Mango fruit juice can increase its protection effect to histological structure of renal cell therefore it can’t give like histological structure in control group. Tujuan Penelitian: Penelitian ini bertujuan untuk mengetahui efek pemberian jus Mangga dan peningkatan dosis jus Mangga terhadap kerusakan struktur histologis sel ginjal mencit yang dipapar parasetamol. Metode Penelitian: Penelitian ini bersifat eksperimental laboratorik dengan Randomized Controlled Trial (RCT). Sampel berupa mencit (Mus musculus) jantan, galur Swiss webster berumur 2-3 bulan dengan berat badan + 20 g. Sampel sebanyak 28 mencit dibagi dalam 4 kelompok, masing-masing kelompok terdiri dari 7 mencit. Teknik sampling yang dipakai adalah incidental sampling. Jus Mangga diberikan selama 14 hari berturut-turut, sedangkan parasetamol dosis toksik diberikan pada hari ke-12, 13, dan 14. Kelompok Kontrol negatif (KKn), mencit diberi akuades saja. Kelompok Kontrol positif (KKp), mencit diberi parasetamol dosis toksik. Kelompok Perlakuan 1 (KP1), mencit diberi jus Mangga dosis I dan parasetamol dosis toksik. Kelompok Perlakuan 2 (KP2), mencit diberi jus Mangga dosis II dan parasetamol dosis toksik. Hari ke-15, mencit dikorbankan dengan cara dislokasi leher, kemudian organ ginjalnya diambil dan dibuat preparat dengan metode blok parafin dan pengecatan Hematoksilin Eosin (HE). Kerusakan sel ginjal dinilai berdasarkan jumlah sel yang mengalami piknosis, karioreksis, dan kariolisis. Data dianalisis menggunakan uji One-Way ANOVA (α = 0,05) dan dilanjutkan dengan uji Least Significant Differences (LSD) (α = 0,05). Hasil Penelitian: Hasil analisis uji One-Way ANOVA menunjukkan adanya perbedaan yang bermakna mengenai kerusakan sel ginjal di antara seluruh kelompok. Hasil analisis uji LSD menunjukkan perbedaan bermakna mengenai kerusakan sel ginjal pada setiap pasang kelompok. Simpulan Penelitian: Pemberian jus Mangga dapat mengurangi kerusakan histologis sel ginjal mencit yang diinduksi parasetamol dan peningkatan dosis jus Mangga dapat meningkatkan efek proteksinya terhadap kerusakan histologis sel ginjal mencit meskipun belum dapat mencapai keadaan seperti pada kelompok kontrol.
