    Abstract

    Background: Andrographis paniculata Ness contains flavonoids, beta-sitosterol, and andrographolide which can lower LDL cholesterol levels of blood. The objective of this research was to determine the effect of Andrographis paniculata Ness leaves extract against blood LDL cholesterol levels of white rats (Rattus norvegicus). Methods: This study was a laboratory experimental research design, used the posttest only controlled group design. Subjects of research were Rattus norvegicus, Strain Wistar, male, 3 months, 200 grams. By incidental sampling technique, data were collected from 25 Rattus norvegicus. Those Rattus norvegicus divided into 5 groups, each group consists of 5 Rattus norvegicus. The negative control group (KKn) received distilled water, the positive control group (KKp) and the 1st-3rd treatment group (KP1-KP3) received high cholesterol diet using yolk egg (6,25 g//kg BW/day. The 1st-3rd treatment group (KP1-KP3) not only received high cholesterol diet but also received multilevel doses of Andrographis paniculata Ness leaves extract (160 mg/200 g BW, 320 mg/200 g BW, and 640 mg/200 g BW). This research was held during 28 days. Data of blood LDL cholesterol levels were collected 10-12 hours after the last treatment. The result was analyzed by using One-Way ANOVA test (α = 0.05) and Posthoc Multiple Comparisons (LSD) test (α = 0.05). Results: The results of One-Way ANOVA test showed a significant difference among five groups p = 0.000 (p < 0.05). The results of LSD test showed significant differences between KKn-KKp, KKn-KP1, KKn-KP2, KKn-KP3, KKp- KP1, KKp-KP2, KKp-KP3, (p < 0.005) and non-significant difference between KP1-KP2, KP1-KP3, and KP2-KP3 (p > 0.005). Conclusion Research: Andrographis paniculata Ness leaves extract can lower blood LDL cholesterol levels on Rattus norvegicus but increasing the dose does not increase its function in lowering blood LDL cholesterol. Keywords: Andrographis paniculata Ness Leaves extract, LDL cholesterol Latar Belakang: Daun Sambiloto mengandung flavonoid, beta-sitosterol, dan andrografolid yang mampu menurunkan kadar kolesterol LDL darah. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak Daun Sambiloto terhadap kadar kolesterol LDL darah tikus putih (Rattus norvegicus). Metode Penelitian: Penelitian ini bersifat eksperimental laboratorik dengan the posttest only controlled group design. Tikus putih jantan galur Swiss Wistar berumur 3 bulan dengan berat badan 200 g diambil secara incidental sampling sebanyak 25 ekor dan dibagi dalam 5 kelompok secara random. Kelompok Kontrol Negatif (KKn) diberi aquades, sedangkan Kelompok Kontrol Positif (KKp) dan Kelompok Perlakuan 1-3 (KP1-3) diberi kuning telur (6,25 g/kg BB/hari). KP1-3 juga diberi ekstrak Daun Sambiloto dengan dosis bertingkat, yaitu 160 mg/200 g BB tikus putih, 320 mg/200 g BB tikus putih, dan 640 mg/200 g BB tikus putih. Perlakuan ini dilakukan selama 28 hari. Pengambilan data kadar kolesterol LDL darah tikus putih dilakukan 10-12 jam setelah perlakuan terakhir. Hasil yang diperoleh, kemudian dianalisis dengan uji One-Way ANOVA (α = 0.05)) yang dilanjutkan dengan Posthoc Multiple Comparisons test (LSD) (α = 0.05). Hasil Penelitian: Hasil uji One-Way ANOVA menunjukkan perbedaan yang signifikan di antara kelima kelompok penelitian p = 0.000 (p < 0.05). Hasil uji LSD menunjukkan perbedaan yang bermakna antara KKn-KKp, KKn-KP1, KKn- KP2, KKn-KP3, KKp-KP1, KKp-KP2, KKp-KP3, (p < 0.05) dan perbedaan yang tidak bermakna antara KP1-KP2-KP3 dan KP2-KP3 (p > 0.05). Simpulan Penelitian: Pemberian ekstrak Daun Sambiloto dapat menurunkan kadar kolesterol LDL darah tikus putih (Rattus norvegicus) tetapi peningkatan dosis tidak mampu meningkatkan fungsinya dalam menurunkan kadar kolesterol LDL darah.
