    Abstract

    MOLECULAR MARKER IDENTIFICATION OF SOME BROWN PLANT HOPPER RESISTANT RICE CULTIVARS BASED ON TOTAL PROTEIN PATTERN. Thesis-S1: Mei Nazilatun N (H0709069). Advisers: Supriyadi, Parjanto. Study Program: Agrotechnology, Faculty of Agriculture, University of Sebelas Maret (UNS) Surakarta. Rice is an important food crop for the people of Indonesia. Brown plant hopper is one of the key pest in rice plant. Control of the pest by using the leafhopper resistant varieties could induce a new brown plant hopper biotype. However, the relation and the resistant cultivar respond to against the brown plant hopper have not revealed yet. The expression of plant respond to against brown plant hopper may be seen on the molecular aspects, namely the expression of genes in the form of total protein. Therefore, there is an idea to identify the respond of the resistant cultivar to against brown plant hopper by using protein pattern. This study aims to determine the respond of the resistant cultivar to against brown plant hopper (N. lugens) by using protein pattern. The research was conducted in October 2012 to December 2012 that took place in the Laboratory of Plant Pests and Diseases of the Faculty of Agriculture, University of Sebelas Maret (UNS) Surakarta and Laboratorim of Microbiology II, Inter-University (PAU), Gadjah Mada University in Yogyakarta. This study includes the planting of susceptible rice cultivars (Cisadane) and resistant cultivars (IR26, IR42 and Inpari 13) with two conditions, that are the brown plant hopper infested and not infested by brown plant hopper. Rice leaf samples were taken for analysis of molecular markers based on total protein by SDS-PAGE method. Electrophoretic protein banding pattern produced a series of proteins that are presented in the form of zimogram. Then tabulated quantitatively with the emergence of bands 1 and the lack of value is 0 to generate a pop-up with the program NTSYS dendogram UPGMA method. The result shows that there is a protein profile pattern difference between rice plant that infested and non-infested by brown plant hopper, and also between resistant cultivars and susceptible cultivar, especially in 50 kDa protein band. The 50 kDa protein band of rice cultivars that are infested by brown plant hopper is different clearly from the non-infested rice cultivars protein band. Therefore, 50 kDa protein band is potential to be used as an identifying mark of brown plant hopper attack and also as tenacity level mark. Hama wereng coklat merupakan salah satu hama penting tanaman padi. Pengendalian hama yang dilakukan dengan pemanfaatan varites unggul tahan wereng mengakibatkan munculnya biotipe baru wereng coklat. Tetapi, hubungan dan respon dari kultivar tahan melawan hama wereng coklat masih belum terungkap. Ekspresi respon tanaman terhadap serangan wereng coklat mungkin terlihat pada aspek molekuler, yakni ekspresi gen dalam bentuk protein total. Oleh karena itu, muncul suatu pemikiran untuk mengidentifikasi profil protein total dalam beberapa kultivar tanaman padi. Penelitian ini bertujuan untuk mengetahui respon kultivar tahan tanaman padi melawan serangan hama wereng coklat berdasarkan marka protein. Penelitian ini dilaksanakan pada bulan Oktober 2012 sampai dengan bulan Desember 2012 yang bertempat di Laboratorium Hama dan Penyakit Tanaman Fakultas Pertanian Universitas Sebelas Maret Surakarta dan Laboratorim Mikrobiologi II, Pusat Antar Universitas (PAU), Universitas Gadjah Mada Yogyakarta. Penelitian ini meliputi penanaman padi kultivar rentan (Cisadane) dan kultivar tahan (IR26, IR42 dan Inpari 13) dengan dua ketentuan yaitu diinvestasi wereng coklat dan tidak diinvestasi wereng coklat. Sampel daun padi diambil untuk dianalisis molekuler berdasarkan marka protein total dengan metode SDS-PAGE. Elektroforesis protein menghasilkan rangkaian pola pita protein yang disajikan dalam bentuk zimogram. Kemudian ditabulasi secara kuantitatif dengan kemunculan pita bernilai 1 dan ketidak munculan bernilai 0 untuk menghasilkan dendogram dengan program NTSYS metode UPGMA. Hasil penelitian menunjukkan bahwa terdapat perbedaan profil pita protein antara tanaman padi yang terserang wereng dan tidak, serta antara kultivar tahan dan rentan, khususnya pada pita protein 50 kDa. Ketebalan pita protein 50 kDa pada tanaman padi yang terinfestasi wereng berbeda jelas dengan profil pita tanaman padi tidak terserang. Oleh karena itu, pita protein total 50 kDa tersebut potensial untuk digunakan sebagai penanda adanya serangan wereng coklat pada tanaman padi dan penanda tanaman tahan atau rentan terhadap serangan wereng.
