    Abstract

    Taro (Colocasia esculenta (L.) Schott) is the plant of food that can be used as the source of carbohydrate. The aim of this research was to study the diversity and relationship Taro in Surakarta region based on the morpholigical, anatomical and isozyme banding pattern characters. The samples were taken using random purposive sampling technique. Twenty samples plant of Taro were taken in district of Surakarta, the are Boyolali, Klaten, Karanganyar, Wonogiri, Sragen, Sukoharjo and Surakarta. The research was conducted on the observation of morphological characters (roots, plant height, leaves), leaves anatomical (stomata, epidermis, mesophyll and palisade) and analysis of isozyme banding pattern by electrophoresis esterase and peroxidase isozyme. The diversity data of morphological, anatomical and isozyme banding patterns that have been analyzed descriptively. the binary data used to calculate the similarity index (IS) between individuals taro. For relationship done by clustering analysis using the program Numerical Taxonomy and Multivariate Analysis System (NTSYSpc) version 2.10 to obtain the relationship dendogram. The result showed the diversity of morphological in midrib ratio (<20,35 and >20,35), petiole color (purplish and not purple) and petiole ratio (<47,6 and >47,6). The diversity anatomical in stomatal index (<15,95 dan >15,95), thick of mesophyll (<294,28 µm and >294,28 µm), length of palisade (<80,81 µm and >80,81 µm), thick of epidermis (<26,71 µm and >26,71 µm) and palisade ratio (<8,45 and >8,45). On the diversity os characters banding pattern is show at 12 esterase banding pattern and 8 peroxydase banding pattern. Based on all the characters of the analyzed, the closest relationships were showed in taro Sukoharjo and Surakarta that had 96% similarity coefficient while taro Klaten 3, Karanganyar 4 and Klaten 4 clustered separately from the another taro with similarity coefficient 76%. Talas (Colocasia esculenta (L.) Schott) merupakan tanaman pangan yang dapat digunakan sebagai sumber karbohidrat. Tujuan dari penelitian ini adalah mengetahui keragaman dan jauh dekatnya hubungan kekerabatan talas di wilayah eks-karesidenan Surakarta berdasarkan karakter morfologi, anatomi dan pola pita isozim. Pengambilan sampel dikoleksi dengan cara purposive random sampling. Sebanyak 20 tanaman talas di wilayah eks-karesidenan Surakarta, meliputi Boyolali, Klaten, Karnganyar, Wonogiri, Sragen, Sukoharjo dan Surakarta. Penelitian yang dilakukan meliputi pengamatan karakter morfologi (akar, tinggi tanaman, daun), anatomi daun (stomata, epidermis, mesofil dan palisade) dan analisis pola pita isozim dengan elektroforesis menggunakan dua sistem isozim yaitu esterase dan peroksidase. Penentuan hubungan kekerabatan dimulai dengan menghitung indeks similaritas (IS) antarindividu talas. Selanjutnya dilakukan analisis pengelompokan (clustering analysis) menggunakan program Numerical Taxonomy and Multivariate Analysis System (NTSYSpc) versi 2.10. Hasil penelitian menunjukkan keragaman morfologi wilayah Eks-karesidenan Surakata berupa rasio pelepah (<20,35 dan >20,35), warna tangkai daun (keunguan dan tidak ungu) dan rasio tangkai daun (<47,6 dan >47,6). Keragaman karakter anatomi berupa indeks stomata (<15,95 dan >15,95), tebal mesofil (<294,28 µm dan >294,28 µm), panjang palisade (<80,81 µm dan >80,81 µm), tebal epidermis (<26,71 µm dan >26,71 µm) dan rasio palisade (<8,45 dan >8,45). Pada keragaman pola pita isozim, isozim esterase memiliki 12 pola pita dan pada isozim peroksidase memiliki 8 pola pita. Berdasarkan semua karakter yang dianalisis ternyata talas Sukoharjo dan Surakarta memiliki hubungan kekerabatan terdekat dengan koefisien kemiripan 96% sedangkan talas Klaten 3, Karanganyar 4 dan Klaten 4 mengelompok tersendiri pada koefisien 76%.
