    Abstract

    Elanda Rahmat Arifyanto, G.0009070, 2012. The Hepatoprotector Effect of Soybean Tempeh Extract in Mice (Mus musculus) Induced by Paracetamol. Mini Thesis, Faculty of Medicine, Sebelas Maret University, Surakarta. Background: Soybean tempeh is known to contain various kinds of antioxidants such as vitamin C, vitamin E, vitamin B2, saponins, isoflavones, phytic acid, lecithin, iron, and phytosterol. Antioxidants are thought to be hepatoprotective. The objectives of this research are to know the influence of soybean tempeh extract to the liver histological damage of mice (Mus musculus) which is induced by Paracetamol and the increase of soybean tempeh extract dose can also increase protection effect to the liver histological damage of mice (Mus musculus) which is induced by Paracetamol. Methods: This was laboratory experimental research with the post test only controlled group design. Samples were 32 male mice, Swiss Webster type, 2-3 months old age and ± 20 g of each weight. Samples were divided into 4 groups of 8 mice each. Sampling technique used in this research was incidental sampling. Mice for control group (K) and the first treatment group (P1) will be given aquadest for 14 days in a row. The second treatment group (P2) and third treatment group (P3) will be given soybean tempeh extracts dose for 14 days in a row. Soybean tempeh extracts dose in each group is 2,2 mg/20 g weight of mice and 4,4 mg/20 g weight of mice. Paracetamol will be given to P1, P2, and P3, with dose 5 mg/20 g weight of mice in 0,1ml aquadest on the day 12, 13, and 14. Finally on day 15th, mice are sacrificed with neck dislocation. After that, we made preparate from the liver that painted by Hematoxillin Eosin. Liver histological is observed and scored base on quantifying of liver histological damaging on pyknosis, karyorhexis, and karyolysis. Data are analized by One- Way ANOVA test (α = 0,05), and continued by Post Hoc Multiple Comparisons test (LSD) (α = 0,05). Results: Result of One-Way ANOVA shows that there was a significant of degree between 4 groups with p = 0,000 (p < 0,05). Result of LSD method there was a significant of degree between K-P1, K-P2, K-P3, P1-P2, P1-P3, and P2-P3 groups with Beach p = 0,000 (p < 0,05). Conclusion: The feeding of soybean tempeh extracts was able to decrease the liver cell damaging of mice and the increase of soybean tempeh extracts dose followed by the increase of protection effect to the liver cell damaging of mice which is induced by paracetamol. Latar Belakang: Tempe kedelai diketahui mengandung berbagai macam antioksidan seperti: vitamin C, vitamin E, vitamin B2, saponin, isoflavon, asam fitat, lesitin, besi, dan fitosterol. Antioksidan tersebut diduga bersifat hepatoprotektif. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak tempe kedelai terhadap kerusakan histologis sel hepar mencit (Mus musculus) yang diinduksi parasetamol dan peningkatan dosis ekstrak tempe kedelai dapat meningkatkan efek proteksinya terhadap kerusakan histologis sel hepar mencit (Mus musculus) yang diinduksi parasetamol. Metode Penelitian: Penelitian ini bersifat eksperimental laboratorik dengan the post test only controlled group design. Sampel berupa mencit jantan, galur Swiss Webster berumur 2-3 bulan dengan berat badan ± 20 gr. Sampel sebanyak 32 ekor mencit dibagi dalam 4 kelompok, masing-masing kelompok terdiri dari 8 ekor mencit. Teknik sampling yang dipakai adalah incidental sampling. Kelompok kontrol (K) dan kelompok perlakuan 1 (P1), mencit diberi aquades selama 14 hari sebagai plasebo. Kelompok perlakuan 2 (P2) dan kelompok perlakuan 3 (P3), mencit diberi ekstrak tempe kedelai selama 14 hari dengan dosis masing-masing 2,2 mg/20 gr BB mencit dan 4,4 mg/20 gr BB mencit. Parasetamol dosis 5 mg/20 gr BB mencit dalam 0,1 ml aquades diberikan pada kelompok P1, P2, dan P3 pada hari ke-12, 13, dan 14. Hari ke-15, mencit dikorbankan kemudian hepar mencit dibuat preparat dengan metode blok parafin dan pengecatan Hematoksilin Eosin (HE). Gambaran histologis hepar diamati dan dinilai berdasarkan jumlah kerusakan histologis yang berupa penjumlahan inti piknosis, karioreksis, dan kariolisis. Data dianalisis dengan menggunakan uji One-Way ANOVA (α = 0,05) dan dilanjutkan dengan uji Post Hoc Multiple Comparisons (LSD) (α = 0,05). Hasil Penelitian: Hasil uji One-Way ANOVA menunjukkan adanya perbedaan yang bermakna antara keempat kelompok dengan p = 0,000 (p < 0,05). Hasil uji LSD menunjukkan adanya perbedaan yang bermakna antara K-P1, K-P2, K-P3, P1-P2, P1-P3, dan P2-P3 dengan masing-masing p = 0,000 (p < 0,05). Simpulan Penelitian: Pemberian ekstrak tempe kedelai dapat mengurangi kerusakan sel hepar mencit (Mus musculus) yang diinduksi parasetamol dan peningkatan dosis ekstrak tempe kedelai dapat meningkatkan efek proteksinya terhadap kerusakan sel hepar mencit.
