    Abstract

    ABSTRACT Insomnia problem often occurs, in both young and old ages and it frequently occurs along with emotional disorders such as anxiety, restlessness, depression or fear. Pegagan extract (Centella asiatica) could exert sedative effect; For that reason, there should be a sedative effect examination on pegagan ethyl acetate fraction on balb/c mice based on sleeping duration parameter. Phytochemical screening was carried out with preliminary and TLC test. This study was experimental research with post-test only control group design. The tested animals used were 15 male balb/c mice, divided randomly into 5 groups. They were positive control (Phenobarbital 6 mg/kgBW), negative control (Na CMC 0,5%), and pegagan ethyl acetate fraction at 0.7 mg/grBW (P1), 1.6 mg/grBW (P2), and 3.2 mg/grBW (P3) doses was given orally. The observation was conducted by recording the mice’s sleeping duration. The data was analyzed with SPSS 16.0 for windows. The result of screening showed that positively contained triterpenoid compound in preliminary test. TLC test showed that Rf values of pegagan ethanol extract (0,78) and pegagan ethyl acetate fraction (0,77) positively contained triterpenoid with Liebermann-Burchad showed green colour. Pegagan ethyl acetate fraction at various doses could exert sedative effect on mice but with significant difference (Sig 0.000<0.05) compared with luminal. It could be found that the higher dose provided longer sleeping duration. Dose of 3.2 mg/grBW provided maximum sleeping duration. Keywords: Centella asiatica (L.)
